What you will do

• Develop novel multi-specific biologics formats for therapeutic-area drug discovery programs.
• Generate and screen stable CHO cell lines using multiple vectors and selection strategies.
• Optimize chain ratios for three- and four-chain molecules and develop batch and fed-batch expression processes.
• Apply targeted integration, FACS-based single-cell sorting, automation, and high-throughput workflows.
• Characterize cell line copy number, mRNA transcript levels, titer, monomer content, and chain-pairing efficiency using ddPCR, analytical HPLC, and related tools.
• Analyze, document, and report experimental results while troubleshooting expression, stability, glycosylation, aggregation, and chain-pairing issues.

Requirements

• Bachelor’s or master’s degree in biology, chemistry, biochemistry, or a related field.
• Typically 5+ years of experience with a bachelor’s degree or 2+ years with a master’s degree.
• Hands-on experience with CHO cell culture and expression of monoclonal, bi-specific, and multi-specific antibodies, including thawing, culturing, expression, and cryopreservation.
• Experience generating stable CHO cell lines that express complex multi-specific molecules.
• Experience with process development, process optimization, and automation.
• Must work onsite in North Chicago, Illinois, United States.

Nice to have

• Analytical characterization of multi-specific biologics.
• Targeted integration, FACS, ddPCR, and vector design experience.
• Experience with Wave bioreactors and mammalian expression.
• Experience resolving poor chain pairing, low expression, aggregation, and glycosylation issues.
