What you will do

• Design, execute, and optimize CRISPR/Cas9, prime editing, and base editing strategies, including multiplex editing across multiple genomic targets.
• Generate and validate guide RNAs, pegRNAs, donor templates, expression constructs, and other genome editing reagents.
• Develop and troubleshoot editing approaches for non-model mammalian systems, improving efficiency, specificity, reproducibility, and scalability.
• Design genotyping assays and analyze Sanger and short- and long-read NGS data to identify intended, unintended, and complex editing outcomes.
• Collaborate with Cell Engineering scientists on reagent transfer, experimental design, editing readouts, and workflow optimization.
• Plan experiments, document results, maintain reagent records, and communicate findings through reports, presentations, and team meetings.

Requirements

• On-site work in Dallas, Texas is required.
• PhD in genetics, molecular biology, genome engineering, synthetic biology, or a related field.
• Hands-on mammalian CRISPR/Cas9 experience and expertise in at least one advanced modality: prime editing or base editing.
• Experience with multiplex genome editing, complex editing outcomes, molecular genetics, DNA repair mechanisms, cloning, DNA assembly, and reagent preparation.
• Ability to independently design genotyping assays and analyze sequencing data with bioinformatics tools and pipelines.
• Ability to work independently from the outset, design experiments, troubleshoot technically complex problems, and collaborate effectively.

Nice to have

• Experience with both prime editing and base editing, including editor and guide optimization.
• Experience with HDR donor design, large insertions, integrase or recombinase-based approaches, and complex genomic modifications.
• Experience with in vitro transcription, RNA purification, reagent quality control, off-target analysis, structural variants, copy number, and allele phasing.
• Experience developing reproducible genome editing analysis pipelines and computational workflows.
