What you will do

• Investigate how post-translational modifications in antibody CDRs affect antigen binding and molecular stability.
• Produce and characterize modified and unmodified mAb/Fab samples using mass spectrometry, SEC, DSC, and binding assays.
• Perform 2D-NMR analysis, including chemical-shift perturbation mapping and identification of PTM-induced structural signatures.
• Produce samples through cell-free expression and design, produce, and evaluate mutated Fab constructs.
• Characterize Fab–antigen interactions using NMR, cryo-EM, crystallography, and HDX-MS to build an integrated structural view.
• Present findings to cross-functional teams and contribute to scientific publications.

Requirements

• PhD in NMR spectroscopy, structural biology, biophysics, or a related field.
• Hands-on protein production experience, including U-13C, 15N, and 2H isotope labelling and specific methyl labelling.
• Experience assigning NMR spectra of high-molecular-weight proteins, including backbone and methyl groups.
• Fluent English and intermediate French are required.
• Strong analytical mindset, curiosity, autonomy, and ability to collaborate in a multidisciplinary team.
