Principal Scientist R&D & cGMP Preparative Chromatography (Preparative HPLC/SFC)

Location: Boston, MA; on-site five days per week with ad hoc flexibility

Salary: $138,000–$207,000 base salary per year, plus eligibility for annual bonus and equity awards.

What you will do

• Lead development, optimization, and scale-up of preparative HPLC and SFC purification methods from semi-preparative to kilogram scale in R&D and cGMP environments.
• Execute and troubleshoot mass-directed and preparative purification campaigns for complex small-molecule drug substances.
• Establish kilo-lab chromatography infrastructure, including equipment procurement, column packing, system qualification, and maintenance.
• Perform multi-step synthesis of drug substance intermediates and APIs at gram-to-kilogram scale, including reactor operations, workups, crystallization, filtration, and drying.
• Build peptide and oligonucleotide synthesis and purification capabilities, including IP-RP HPLC, AEX/CEX, ultrafiltration, and diafiltration.
• Lead cross-functional technical initiatives, mentor scientific staff, and own GMP documentation, quality, safety, and audit readiness.

Requirements

• 10+ years of industry experience with a bachelor's degree, 7+ years with a master's degree, or 4+ years with a Ph.D. in a relevant discipline.
• Expertise in preparative HPLC, SFC, IP-RP, AEX, CEX, SMB, and SEC, including method development, column selection, mobile-phase optimization, and mass-directed purification.
• Hands-on experience with cGMP pharmaceutical separations, drug substance development, manufacturing, documentation, and quality systems.
• Ability to perform kilogram-scale synthetic organic chemistry and interpret HPLC, LC-MS, NMR, and Karl Fischer data.
• Experience leading cross-functional projects, communicating complex technical findings, and mentoring junior scientists and technical staff.
• Ability to work on-site in Boston, MA five days per week.

Nice to have

• Ph.D. in analytical chemistry, organic chemistry, medicinal chemistry, chemical engineering, or a related discipline.
• Experience with Empower or OpenLab chromatography data systems.
• Experience with solid-phase phosphoramidite chemistry and kilogram-scale oligonucleotide purification.
