Senior Research Scientist (CRISPR)

What you will do

• Design and execute pooled CRISPR knockout, activation, interference, and base-editing screens.
• Manage custom library design, cloning, virus production, transduction, and screening across iPSCs, primary cells, and other complex models.
• Lead iPSC maintenance, expansion, 2D and 3D culture, and differentiation into neurons, cardiomyocytes, immune cells, and other specialized lineages.
• Develop scalable cell-to-data workflows integrating automated liquid handling with high-throughput sequencing and imaging readouts.
• Collaborate with investigators and computational biologists to interpret results, validate hits, and conduct secondary and arrayed assays.
• Mentor Research Associates and oversee laboratory logistics, reagent scaling, equipment maintenance, and safety compliance.

Requirements

• PhD in Molecular Biology, Genetics, Bioengineering, or a related field with 5+ years of postdoctoral or industry experience, or a Master’s degree with 8+ years of specialized experience.
• Mandatory hands-on experience with pooled CRISPR screening, including library cloning, lentiviral production, and screen execution.
• Deep experience with iPSC culture and differentiation, including long-term 2D and 3D cultures.
• Expertise in molecular cloning, NGS library preparation, and qPCR or ddPCR for library quantification.
• Ability to use basic bioinformatics tools for gRNA design and screening data quality control and collaborate on complex hit analysis.
• Ability to work on-site in Burlington, Massachusetts.

Nice to have

• Experience with liquid handling platforms such as Hamilton, Beckman, or Agilent.
• Experience integrating automation into high-throughput screening workflows.
